چکیده

Abstract The intestinal microbiota contributes to gut immune homeostasis, where short-chain fatty acids (SCFAs) function as the major mediators. We aimed elucidate immunomodulatory effects of acetate, propionate, and butyrate. With that in mind, we sought characterise expression SCFA receptors transporters well SCFAs’ impact on activation different cells. Whereas all three SCFAs decreased tumour necrosis factor (TNF)-α production activated T cells, only butyrate propionate inhibited interferon (IFN)-γ, interleukin (IL)-17, IL-13, IL-10 production. Butyrate chemokine CCR9 CCR10 T- B-cells, respectively. Similarly, were effective inhibitors IL-1β, IL-6, TNF-α, myeloid cells upon lipopolysaccharide R848 stimulation. Acetate was less efficient at inhibiting cytokine except for IFN-α. Moreover, IL-6 TNF-α monocytes, dendritic (mDC), plasmacytoid (pDC), whereas acetate relatively more prominent pDCs. In monocytes mDCs, a inhibitor, but it equally pDCs activation. also studied ability induce trained immunity or tolerance. – not prevented Toll-like receptor-mediated SCFA-trained demonstrated by reduced TNF-α. Our findings indicate are adaptive innate response did immunity. may be explained differential receptor transporter profiles

Fibroblast and T cells conditioned media induce maturation dendritic cell and promote T helper immune response

Dendritic cells (DCs) induce pathogen-specific T cell responses. We comprehensively studied the effects of addition of maturation stimulus, fibroblasts (fibroblast conditioned medium), PHA activated T cells (T cell conditioned medium), and mixture of fibroblast & PHA activated T cells (FCM-TCCM) conditioned media on maturation of DCs. Monocytes were cultured with GM-CSF and IL-4 for five days. ...

TIM-3 regulates innate immune cells to induce fetomaternal tolerance.

TIM-3 is constitutively expressed on subsets of macrophages and dendritic cells. Its expression on other cells of the innate immune system and its role in fetomaternal tolerance has not yet been explored. In this study, we investigate the role of TIM-3-expressing innate immune cells in the regulation of tolerance at the fetomaternal interface (FMI) using an allogeneic mouse model of pregnancy. ...

Short chain fatty acids and colonic health.

Recently, colonic health has been linked to the maintaining overall health status and reducing the risk of diseases by changes in lifestyle. Functional foods, such as "prebiotics" and "probiotics", dietary fibers, and other dietary components that target the colon and affect its environment enhancing short fatty acid (SCFA) production have been at the forefront. The topic of this review is the ...

Short-chain fatty acids inhibit intestinal trefoil factor gene expression in colon cancer cells.

Intestinal trefoil factor (ITF) gene expression was detected in five colon cancer cell lines. ITF was synthesized by mucous cells of LIM 1215 and LIM 1863 lines, from which it is secreted constitutively. The ITF mRNA transcript was estimated to be 0.6 kb. In LIM 1215 cells, the expression of ITF was potently and dose-dependently inhibited by short-chain fatty acids (butyrate > propionate > acet...
